摘要(Abstract):
本文用高效液相色谱法测定人血浆中布洛芬的浓度,以消炎痛为内标,用正已烷:氯仿:异丙醇(65:25:10)混合溶剂提取,以Zorbax—C_8柱作固定相,以甲醇:0.1mol/L醋酸缓冲液(pH=4.7):0.085%的磷酸溶液(135:35:30)为流动相,225nm为检测波长,用峰高比定量。线性范围为0.5~40μg/ml,最低检测浓度为0.1μg/ml.用本法测定了健康人口服布洛芬片后的血药浓度,进行了药代动力学参数计算。
A highly sensitive and specifically analytical HPLC method for determining ibuprofen level in plasma was established. The chromatographic conditions were: a ZorbaxC_8(5μm)reversed phase column(2.5×4.6); amethanol-0.1mol/L acetate buffer(pH4.7)-0.085%H_3PO_4(135:35:30) mobile phase; UV detecter at 225nm. The mean recovery of ibuprofen within the concentration range was 92.7%. The good linear range was 0.539.8μg/ml. The pharmacokinetics of ibuprofen is discussed in this paper.
