基于生物信息学分析高表达RAI14对结肠癌及其耐药性的影响The influence of RAI14 upregulation on colon cancer and drug resistance based on bioinformatic analyse

摘要(Abstract):
目的 研究维甲酸诱导蛋白14(retinoic acid induced 14,RAI14)与结肠癌及其耐药性之间的关系。方法 运用Oncomine、UALCAN、HPA数据库分析RAI14在正常结肠组织与结肠癌组织中的表达差异;PrognoScan联合HPA数据库分析RAI14表达量与结肠癌患者预后的相关性;GEO数据库分析RAI14在结肠癌化疗耐药性细胞系中的表达量;CTRP数据库分析大肠肿瘤细胞系RAI14表达量与化学小分子敏感性的相关性;TargetScan联合UCSC Xena分析RAI14与hsa-miR-183的相关性;GEO数据库分析hsa-miR-183在结肠癌和耐药性结肠癌细胞系中的表达;COREMINE分析RAI14、hsa-miR-183、结肠癌、耐药性及其生物过程的相关性。结果 RAI14在结肠癌组织中高表达,而且与结肠癌患者不良预后显著相关。化疗耐药的结肠癌细胞系中RAI14表达量也显著高于对化疗药物敏感的结肠癌细胞系,RAI14基因表达量与大肠肿瘤细胞系对化学小分子的敏感性负相关。hsa-miR-183靶向于RAI14保守序列,并且结肠癌组织的MIR183基因拷贝量与RAI14表达量负相关。hsa-miR-183在结肠癌及耐药性细胞系中低表达,文本挖掘显示hsa-miR-183与结肠癌耐药性显著相关。结论 高表达RAI14与结肠癌及其耐药性显著相关,并且可能通过hsa-miR-183转录后调控实现。
Objective To analyze the association of RAI14 with colon cancer and drug resistance.Methods The Oncomine, UALCAN,and HPA databases were used to detect the mRNA expression of RAI14 in normal tissue and colon cancer.PrognoScan combined with HPA database was used to analyze the correlation of RAI14 expression with prognosis in colon cancer patients.GEO database was used to analyze RAI14 expression in colon cancer drug resistant cell lines.The correlation analysis between RAI14 expression and small molecules sensitivity in colon cancer cell was conduct by CTRP database.Target Scan combined with UCSC Xena were used to analyzed the correlation of RAI14 and hsa-miR-183.GEO database was used to analyzed the expression of hsa-miR-183 in colon cancer and drug resistant cell lines.The correlation among RAI14,hsa-miR-183,colon cancer, drug resistance and their biological process was analyzed with COREMINE.Results RAI14 expression was upregulated in colon cancer and associated with poor prognosis in patients with colon cancer.RAI14 expression was also higher in colon cancer cell lines with drug resistance compared with drug sensitive.RAI14 gene expression level was negatively associated with small molecule sensitivity in large intestine tumor cell lines.hsa-miR-183 targeted to conserved sequence of RAI14,and MIR183 gene copy number was negatively associated with RAI14 gene expression in colon cancer patients.hsa-miR-183 was downregulated in colon cancer and drug resistant cell lines.Text mining showed that hsa-miR-183 was significantly associated with colon cancer drug resistant.Conclusion Overexpression of RAI14 is significantly associated with colon cancer and drug resistance, and possibly regulated by hsa-miR-183.
