摘要(Abstract):
目的建立同时测定大株红景天中红景天苷、酪醇和没食子酸含量的方法。方法采用HPLC法。色谱柱为Inertsil ODS-SP柱(250 mm×4.6 mm,5μm);流动相为乙腈-体积分数0.03%磷酸水溶液(体积比7∶93);检测波长为220 nm;流速为1.0 mL.min-1。结果红景天苷、酪醇和没食子酸质量浓度分别在4.100121.9 mg.L-1(r=0.999 9,n=6)、1.10121.9 mg.L-1(r=0.999 9,n=6)、1.1031.9 mg.L-1(r=0.999 8,n=6)、2.0031.9 mg.L-1(r=0.999 8,n=6)、2.0060.7 mg.L-1(r=0.999 9,n=6)内与峰面积呈良好的线性关系。平均回收率(n=9)分别为99.1%、98.9%和103.0%,RSD分别为3.0%、2.1%和0.8%。结论本方法快捷、简便,结果准确、可靠、重复性好,可作为大株红景天药材中红景天苷、酪醇和没食子酸的测定方法。
Objective To establish a method for the determination of salidroside,tyrosol and gallic acid in Rhodiola kirilowii(Regel) Maxim by HPLC.Methods The chromatographic method was carried out on Inertsil ODS-SP column(250 mm×4.6 mm,5 μm) using acetonitrile-0.03% phosphoric acid(V∶V=7∶93) as the mobile phase;the detection wavelength was set at 220 nm and the flow rate was 1.0 mL·min-1.Results The linear rangers were 4.100-121.9 mg·L-1 for salidroside,1.10-31.9 mg·L-1 for tyrosol and 2.00-60.7 mg·L-1 for gallic acid,respectively.The average recoveries(n=9) were 99.1%,98.9% and 103.0%;RSD(n=9) were 3.0%,2.1% and 0.8%.Conclusions The method is simple,accurate with good reproducibility.It can be used for the quality control of Rhodiola kirilowii(Regel) Maxim.
