摘要(Abstract):
目的建立高效液相色谱法测定蛋氨酸脑啡肽含量。方法采用RP-HPLC法。色谱柱为J.T.Baker C18柱(250 mm×4.6 mm,5μm);流动相A为含体积分数0.1%三氟乙酸的去离子水,流动相B为含体积分数0.1%三氟乙酸的体积分数为60%的乙腈水溶液;流速为1.0 mL.min-1;检测波长为210 nm。结果蛋氨酸脑啡肽质量浓度在7~280 mg.L-1内线性关系良好(r=0.9998),平均回收率为98.54%,RSD为0.89%。结论此方法快捷、精确、重复性好,可用于蛋氨酸脑啡肽的含量测定。
Objective To establish an assay method for methionine enkephalin(MEK) by HPLC.Methods The content of MEK was determined with reversed phase high performance liquid chromatography.The column was J.T.Baker C18(250 mm×4.6 mm,5 μm) and the mobile phases were:Deionized water with 0.1% trifluoroacetic acid for A and 60% acetonitrile with 0.1% trifluoroacetic acid for B.The flow rate of the mobile phase was 1.0 mL min-1.The content was determined at wavelength of 210 nm by UV monitor.Results The standard calibration curves of MEK concentration were linear at the range of 7-280 mg·L-1.Good result was achieved with average recovery of 98.54% and RSD of 0.89%.Conclusions The method is validated for high resolution,linearity,fast,accuracy and repeatability.The experiment provides a method for the analysis and determination of MEK.
