RP-HPLC法测定尿液中非蛋白氮代谢产物的含量

摘要(Abstract):

目的建立RP-HPLC法同时测定尿液中尿素、肌酸、肌酐、尿酸和马尿酸5种非蛋白氮代谢产物。方法色谱柱为Century C18柱(200 mm×4.6 mm,5μm),以乙腈-20 mmol.L-1乙酸铵缓冲液(pH 6.8)为流动相,进行梯度洗脱,检测波长为220 nm。结果肌酸、肌酐、尿酸和马尿酸的质量浓度在2~100 mg.L-1内、尿素的质量浓度在100~1 000 mg.L-1内线性关系良好,方法平均回收率为97.6%~105.7%,RSD均小于4.2%。结论该法灵敏、准确,适用于临床尿液中非蛋白氮代谢产物的检测。
Objective To establish an RP-HPLC method for the simultaneous determination of 5 kinds of nonprotein-nitrogen metabolites in urine(urea,creatine,creatinine,uric acid and hippuric acid).Methods The five compounds were successfully separated on a C18 column(200 mm×4.6 mm,5 μm) using a mobile phase of acetonitrile-ammonium acetate(20 mmol·L-1,pH 6.8) with gradient elution.The detection wavelength was selected at 220 nm.Results The method was linear over the concentration range of 2100 mg·L-1 for creatine,uric acid,creatinine and hippuric acid and 100-1 000 mg·L-1 for urea.The mean recovery of the method was 97.6%-105.7%.The precisions were less than 4.2%.Conclusions The method is sensitive and accurate,and is suitable for determination of nonprotein-nitrogen metabolites in urine.
