RP-HPLC法同时测定金莲花中5种黄酮苷类成分的含量
Simultaneous determination of five flavonoid glycosides in Trollius chinensis Bunge by RP-HPLC

摘要(Abstract):

目的建立反相高效液相色谱法同时测定不同产地金莲花中荭草素-2″-O-β-L-半乳糖苷、荭草苷、牡荆苷、2″-O-(2-甲基丁酰基)牡荆苷和芹菜素-8-C-(2″-O-阿魏酰基)-β-D-葡萄糖共5种有效成分含量的方法。方法采用Thermo C18色谱柱(250 mm×4.60 mm,5μm)进行分离,以乙腈-体积分数0.1%磷酸溶液为流动相进行梯度洗脱,流速为1.0 m L·min-1,检测波长为340 nm,柱温为30℃。结果荭草素-2″-O-β-L-半乳糖苷、荭草苷、牡荆苷、2″-O-(2-甲基丁酰基)牡荆苷和芹菜素-8-C-(2″-O-阿魏酰基)-β-D-葡萄糖分别在质量浓度49.23~492.3 mg·L-1(r=0.999 9)、20.52~205.2 mg·L-1(r=0.999 9)、11.70~117.0 mg·L-1(r=0.999 9)、4.170~41.70 mg·L-1(r=0.999 9)和7.290~72.90 mg·L-1(r=0.999 9)内与峰面积呈良好的线性关系;平均加样回收率分别为99.7%、99.5%、101.3%、99.0%和100.6%(n=6)。不同产地金莲花药材中这5种黄酮苷类成分含量差异较大。结论该方法可用于金莲花药材的质量控制。
Objective To establish an RP-HPLC method for the simultaneous determination of 2″-O-β-Lgalactopyranosylorientin,orientin,vitexin,2″-O-( 2-methylbutyryl) vitexin and apigenin-8-C-( 2″-O-feruloyl)-β-D-glucosidein Trollius chinensis Bunge collected from different regions. Methods The Thermo C18( 250 mm × 4. 60 mm,5 μm) column was used with a gradient elution of acetonitrile-0. 1% phosphoric acid solution at the w avelength of 340 nm. The flow rate w as 1. 0 m L·min-1 and column temperature w as at30 ℃. Results The method w as developed w ith good linearities over the range of 49. 23-492. 3 mg·L-1( r =0. 999 9) for 2″-O-β-L-galactopyranosylorientin,20. 52-205. 2 mg·L-1( r = 0. 999 9) for orientin,11. 70-117. 0 mg·L-1( r = 0. 999 9) for vitexin,4. 170-41. 70 mg·L-1( r = 0. 999 9) for 2″-O-( 2-methylbutyryl)vitexin and 7. 290-72. 90 mg·L-1( r = 0. 999 9) for apigenin-8-C-( 2″-O-feruloyl)-β-D-glucoside,respectively. And the recoveries w ere 99. 7%,99. 5%,101. 3%,99. 0% and 100. 6%,respectively. A significant variability in the contents of five flavonoid glycosides w as found among the tested 23 samples. Conclusions The method is quick,simple and accurate,w hich can be used for quality evaluation of Trollius chinensis Bunge.
