UPLC法同时测定参苓白术散中补骨脂素、异补骨脂素、白术内酯Ⅲ和异欧前胡素的含量Simultaneous determination of psoralen,isopsoralen,atractylenolide Ⅲ and isoimperatorin in Shenling Baizhu powder by UPLC

摘要(Abstract):
目的采用UPLC法,建立参苓白术散中补骨脂素、异补骨脂素、白术内酯Ⅲ和异欧前胡素的含量测定方法。方法采用Universil XB-C8柱(150 mm×2.1mm,1.8μm),流动相为甲醇(A)-体积分数为0.1%的磷酸水溶液(B),进行梯度洗脱,检测波长:254 nm,进样量:5μL,流速:0.2 ml·min~(-1),柱温:35℃。结果补骨脂素、异补骨脂素、白术内酯Ⅲ和异欧前胡素质量浓度的线性分别为2.50~80.00、2.50~80.00、7.50~240.00、2.50~80.00 mg·L~(-1),相关系数分别为0.999 7、0.999 9、0.999 7、0.999 9,平均回收率分别为99.7%、98.5%、97.6%、100.3%,RSD分别为1.0%、0.8%、1.0%、1.2%(n=6)。结论本方法可为参苓白术散的质量控制提供实验依据。
Objective To establish an UPLC method for simultaneous determination of psoralen,isopsorale,atractylenolide III and isoimperatorin in Shenling Baizhu powder( traditional Chinese medicine). Methods The separation was developed on a Universil XB-C8 column( 150 mm × 2. 1 mm,1. 8 μm) by gradient elution with methanol-water( containing 0. 1%( φ) phosphoric acid) as the mobile phase,flowrate at 0. 2 mL·min~(-1),detection wavelength at 254 nm and column temperature at 35 ℃. Results The calibration curve was linear within the range of 2. 50-80. 00 mg·L~(-1)( r = 0. 999 7),2. 50-80. 00 mg·L~(-1)( r =0. 999 9),7. 50-240. 00 mg·L~(-1)( r = 0. 9997),2. 50-80. 00 mg·L~(-1)( r = 0. 9999) for psoralen,isopsoralen,atractylenolide III and isoimperatorin,respectively. The average recoveries were 99. 7%( RSD =1. 0%),98. 5%( RSD = 0. 8%),97. 6%( RSD = 1. 0%) and 100. 3%( RSD = 1. 2%) for psoralen,isopsoralen,atractylenolide III and isoimperatorin,respectively. Conclusion The method can be used for the quality control of Shenling Baizhu powder.
