用Chiralpak IC柱分离4种碱性药物异构体Enantioseparation of four basic drugs by Chiralpak IC column

摘要(Abstract):
目的建立手性HPLC法分离苄氟噻嗪、甲氯噻嗪、卡维地洛和戊乙奎醚4种碱性药物异构体。方法使用Chiralpak IC手性固定相法,通过考察流动相中酸碱添加剂种类、极性醇类改性剂种类、流动相醇烷比例、柱温和流速等因素对异构体分离度的影响,对分离条件进行优化。结果在优化的色谱条件下,4种药物异构体均达到了基线分离。结论以Chiralpak IC柱为手性固定相的HPLC法适于分离上述4种碱性药物的异构体。
Objective To establish high performance liquid chromatography methods for the chiral separation of four basic drugs including bendroflumethiazide,methyclothiazide,carvedilol and penehyclidine hydrochloride. Methods With the application of Chiralpak IC column,the factors affecting separation process such as acidic and basic addictives,the types and ratios of mobile phase,column temperature and flowrate were investigated. Results Under the optimal condition,four basic drugs are completely separated.Conclusion Chiralpak IC column can achieve complete separation of these four basic drugs.
