单组分妥布霉素产生菌的选育Breeding of single component producer of tobramycin

摘要(Abstract):
目的提高妥布霉素产生菌的发酵单位。方法以黑暗链霉菌UVS— 5 1为出发菌株 ,采用UV和NTG为诱变剂 ,结合耐受妥布霉素 ,运用固体循环诱变筛选高产菌株。结果获得两株高产稳定变株NS— 81和NT— 2 6 ,这两株菌的摇瓶发酵效价分别比出发菌株提高 5 3%和 77%。经薄层层析检验 ,该两变株的产物仍为单一的氨甲酰妥布霉素。结论出发菌株经UV和NTG诱变处理后结合耐自身代谢物的驯育 ,正变率显著提高。通过引入循环诱变筛选的思路 ,大大缩短的实验周期 ,有效地提高了诱变育种的工作效率 ,能在较短时间内选育到理想变株 ,因此 ,循环诱变筛选法是一种较好的选育方法。
Objective To improve the titer of the single component producer of tobramycin. Methods UV and NTG were regarded as the mutagens,the tolerant treatment was used, and two mutants were obtained through the cyclic screening of agar disc. Results Two stable mutants were isolated by the above methods. The titers of antibiotic in shaken flasks produced by the two mutants NS 81 and NT 26 were 53% higher than that of the control respectively. They were all identified to be a single component producer of tobramycin by means of TLC. Conclusions Original strain was treated with UV and NTG as well as tolerant treatment, positive mutation frequency was improved obviously; The cycle of the test was shortened widely; the efficiency of mutation breeding was improved effectively and two good mutants were obtained in a short time by the method of cyclic screening of agar disc. So the method of the cyclic screening was considered a good method.
