HPLC法同时测定苦碟子注射液中4种成分的含量

摘要(Abstract):

目的建立同时测定苦碟子注射液中绿原酸、木犀草素-7-O-龙胆二糖苷、木犀草素-7-O-β-D-吡喃葡萄糖苷和木犀草素-7-O-β-D-吡喃葡萄糖醛酸苷含量的HPLC法。方法色谱柱为Agela Pro-mosil C18柱(250 mm×4.6 mm,5μm);流动相为乙腈(A)-体积分数为1.0%乙酸水溶液(B),梯度洗脱;流速为1.0 mL.min-1;检测波长为350 nm。结果绿原酸、木犀草素-7-O-龙胆二糖苷、木犀草素-7-O-β-D-吡喃葡萄糖苷和木犀草素-7-O-β-D-吡喃葡萄糖醛酸苷的线性关系良好,线性范围分别为0.730~5.84 mg.L-1、0.530~4.24 mg.L-1、0.990~7.92 mg.L-1和2.00~16.0 mg.L-1;平均回收率分别为98.8%、99.9%、99.6%和100.6%,RSD分别为0.8%、1.1%、1.0%和0.6%(n=5)。结论该法简便、可行,重现性好,结果准确,为苦碟子注射液质量控制提供参考依据。
Objective To develop a method for the quantitative analysis of chlorogenic acid,luteolin-7-O-gentibioside,luteolin-7-O-β-D-glucopyranoside,luteolin-7-O-β-D-glucuronopyranoside in the injection of Ixeris sonchifolia simultaneously.Methods The injection was determined by Agela Promosil C18 column(250 mm×4.6 mm,5 μm);the mobile phase consisted of acetonitrile(A)and 1.0% HOAc(B)with gradient elution was used;the flow rate was 1.0 mL · min-1.The detector wavelength was set at 350 nm.Results The linear response ranges were in 0.730-5.84 mg · L-1,0.530-4.24 mg · L-1,0.990-7.92 mg · L-1,2.00-16.0 mg · L-1,respectively.The recovery ranges were 98.8%,99.9%,99.6%,100.6%,respectively.The relative standard deviations(RSDs)were 0.8%,1.1%,1.0%,0.6%(n=5),respectively.Conclusions This method can be used for the quality control of the injection of of Ixeris sonchifolia(Bge.)Hance.
