蟾饲五谷虫中华蟾酥毒基的含量测定

摘要(Abstract):

目的建立反相高效液相色谱法测定蟾饲五谷虫中华蟾酥毒基的含量。方法采用Ecosil C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-5 g.L-1磷酸二氢钾溶液(体积比38∶62)(用磷酸调节pH值为3.2)为流动相,检测波长为296 nm,柱温为40℃,流速为1.0 mL.min-1。结果华蟾酥毒基的线性范围为6.0~30.0 mg.L-1(r=0.998 7,n=5),平均回收率为100.0%(RSD=1.54%,n=6)。结论该方法操作简便、结果准确,可用于蟾饲五谷虫的质量控制。
Objective To establish an RP-HPLC method for determination of cinobufagin in maggot bred by toad.Methods A column packed with Ecosil C18 column(250 mm×4.6 mm,5 μm)was used as the stationary phase,the mixture of acetonitrile-KH2PO4(the pH of solution was adjusted to 3.2 by phosphoric acid)(V∶ V=38∶ 62)was served as mobile phase.The detection wavelength was set at 298 nm;the column temperature was 30 ℃;the flow rate was 1.0 mL · min-1.Results The linear range of cinobufagin was 6.0-30.0 mg · L-1(r=0.998 7,n=5),the average recovery of cinobufagin was 100.0%(RSD=1.54%,n=6).Conclusions The method is found to be convenient and accurate,and can be used for the quality control of cinobufagin.
