摘要(Abstract)：
目的建立测定人血浆中头孢克肟浓度的高效液相色谱法。方法色谱柱为ThermoHypersil KeystoneODS 2 ,流动相为乙腈 10mmol·L-1磷酸盐缓冲液 (V∶V =15∶85 ) ,紫外检测波长为2 91nm ,替硝唑为内标。结果头孢克肟浓度线性范围为 0 13～ 8 83μmol·L-1,回归方程Y =0 10 2 3c+0 6 2× 10 -3 ,r =0 9992 ;回收率 :98 3% ,日内和日间RSD分别为 4 6 %和 4 9% ,整个测定时间 12min左右。结论该方法简便、快速、准确 ,适用于临床上头孢克肟药物动力学的研究 ,确保临床用药质量。
Objective To establish a HPLC method to determine the concentrtion of cefixime in the human plasma.Method The assay was conducted on the Thermo Hypersil-Keystone ODS-2 column with acetonitrile- 0.01mol·L -1 PBS (pH=2.6) (15∶85) as the mobile phase.The detection wavelength was set at 291 nm,and the tinidazole was taken as the internal standard.Results The linearity range was 0.13～ 8.83 μmol·L -1 with a regressive equation of Y= 0.102 3c+0.62×10 -3, r=0.999 2.The recovery was 98.5%.RSD: 2.4% (intra-day) and 4.4% (inter-day).Conclusion The method is shown to be convenient,accurate,rapid,and suitable for determination of cefixime in the human plasma and pharmacokinetic studies of cefixime.
