摘要(Abstract):
目的建立HPLC法同时测定山楂叶提取物中牡荆素-2″-O-鼠李糖苷、金丝桃苷、槲皮素、山奈酚的含量。方法色谱柱为DIAMONSIL C18柱(250 mm×4.6 mm,5μm);流动相为甲醇-体积分数为0.5%乙酸水溶液,梯度洗脱[0~15 min,φ(甲醇)=40%~65%;15~20 min,φ(甲醇)=65%~100%;20~40 min,φ(甲醇)=100%];流速为1.0 mL.min-1;波长为365 nm。结果牡荆素-2″-O-鼠李糖苷、金丝桃苷、槲皮素、山奈酚的线性关系良好,其质量浓度分别在14.0~112 mg.L-1、3.50~28.0 mg.L-1、6.25~50.0 mg.L-1、0.550~4.40 mg.L-1内与峰面积呈良好的线性关系。平均回收率分别为101.3%、98.6%、107.0%、98.1%,RSD分别为1.1%、2.2%、2.6%、2.1%(n=5)。结论该法简便、可行,重现性好,结果准确,可作为山楂叶提取物的含量测定方法,为其质量控制提供参考依据。
Objective To develop a method for the quantitative analysis of vitexin-2″-O-rhamnosyl,hyperoside,quercetin,kaempferol in the extract of hawthorn leaves. Methods The extract was determined by DIAMONSIL-C18 column(250 mm×4.6 mm,5 μm).The mobile phase consisted of methanol and HOAc(φ=5%),gradient elution(0-15 min,φ(methanol)=40%-65%;15-20 min,φ(methanol)=65%-100%;20-40 min,φ(methanol)=100%);The flow rate was 1.0 mL·min-1.The DAD detector wavelength was set at 365 nm.The column temperature was ambient temperature. Results The linear response ranges of the four compounds were 14.0-112 mg·L-1,3.50-28.0 mg·L-1,6.25-50.0 mg·L-1,0.550-4.40 mg·L-1,respectively.The recovery range of the four compounds was 101.3%,98.6%,107.0%,98.1%,respectively,and the relative standard deviations(RSDs)were less than 3.0%(n=5). Conclusions This method can be used for the quality control of the extract of hawthorn leaves.
