摘要(Abstract):
目的建立HPLC法测定灯台叶片中灯台树次碱(scholarisine)、19-表灯台树次碱(19-epischolarisine)和瓦莱萨明碱(vallesamine)的含量。方法采用Diamonsil-C_(18)(2)(250 mm×4.6 mm,5μm)色谱柱,流动相:体积分数为0.3%的甲酸水溶液(A),乙腈-甲醇(体积比为:80:20)(B),梯度洗脱,流速:1.0 m L·min~(-1),柱温:30℃,紫外检测波长:285 nm。结果灯台树次碱、19-表灯台树次碱和瓦莱萨明碱的质量浓度分别在3.744~37.44、1.116~11.16、3.840~38.40 mg·L~(-1)内与峰面积呈良好的线性关系(r>0.999 0),平均加样回收率分别为98.9%(RSD=2.2%)、100.9%(RSD=2.6%)和99.9%(RSD=1.5%)(n=6)。结论该方法可用于同时测定灯台叶片中灯台树次碱、19-表灯台树次碱和瓦来萨明碱的含量。
Objective To develop a HPLC method for simultaneous determination of scholarisin(DT4),19-epischolarisine(DT-2) and vallesamine(DT-4) in Dengtaiye tablets.Methods The separation was performed on a Diamonsil-C_(18)(2) column(250 mm×4.6 mm,5μm) with a gradient mobile phase composed of acetonitrile-methanol(8:2)(A) and 0.3%formic acid(B) at the flow rate of 1.0 mL·min~(-1).The column temperature was 30℃ and the detection wavelength was 285 nm.Results The linear range of calibration curves for DT4,DT-2 and DT-4 was 3.744-37.44,1.116-11.16 and 3.840-38.40 mg·L~(-1)(r >0.999 0),respectively.The average recovery of DT4,DT-2 and DT-4 was 98.9%,100.9%and 99.9%with relative standard deviations(RSDs) of 2.2%,2.6%and 1.5%(n = 6),correspondingly.Conclusions This method is selective,accurate,rapid,which can be applied for simultaneous determination of DT-1,DT-2and DT-4 in Dengtaiye tablets.
