加味生化妇康颗粒的HPLC指纹图谱和含量测定HPLC fingerprint and content determination of Jiawei Shenghua Fukang granules

摘要(Abstract):
目的 采用高效液相色谱,结合多元统计分析,建立加味生化妇康颗粒的HPLC指纹图谱,对羟基苯甲酸,香蒲新苷,刺芒柄花苷和甘草酸的含量测定方法。方法 以对羟基苯甲酸为参照,绘制10批样品的指纹图谱,相似度评价,确定共有峰,运用SPSS 19.0、SIMCA 13.0软件进行聚类分析、主成分分析;最后测定4种主要成分的含量。结果 10批样品共有18个共有峰,相似度均大于0.85;共指认了对羟基苯甲酸、香蒲新苷、刺芒柄花苷和甘草酸4个共有峰。结果显示,10批加味生化妇康颗粒样品可聚为3类,3个主成分的累积方差贡献率为91.0%。对羟基苯甲酸,香蒲新苷,刺芒柄花苷和甘草酸质量浓度分别在4.80~48.00、13.14~134.00、3.20~32.00、12.60~126.00 mg·L~(-1)内与峰面积呈良好的线性关系,相关系数分别为0.999 7、0.999 6、0.999 5、0.999 6,平均回收率分别为92.9%、94.3%、96.9%和95.4%;RSD分别为2.7%、2.1%、2.4%和2.8%(n=9)。结论 建立的分析方法结合多元统计分析和含量测定适用于加味生化妇康颗粒质量质量评价。
Objective The HPLC fingerprint of Jiawei Shenghua Fukang granules was established by high performance liquid chromatography combined with multivariate statistical analysis, and the contents of p-hydroxybenzoic acid, typhaneoside, formononetin and glycyrrhizic acid were determined.Methods Taking p-hydroxybenzoic acid as the reference, the fingerprints of 10 batches of samples were established, and the similarity was evaluated to determine the common peak.The contents of four main components were determined by cluster analysis and principal component analysis with SPSS 19.0 and SIMCA 13.0 software.Results There were 18 common peaks whose similarities were greater than 0.85 in these 10 batches of samples, and four common peaks were identified, including p-hydroxybenzoic acid, typhaneoside, anthocyanin and glycyrrhizic acid.The results showed that 10 batches of Jiawei Shenghua Fukang granules samples could be grouped into three categories, and the cumulative variance contribution rate of the three principal components was 90.938%.The mass concentrations of P-hydroxybenzoic acid, typhaneoside, anthocyanin and glycyrrhizic acid showed a good linear relationship with the peak area within 4.80-48.00,13.14-134.00,3.20-32.00 and 12.60-126.00 mg·L~(-1),respectively.The correlation coefficients were 0.999 7,0.999 6,0.999 6 and the average recoveries were 92.9%,94.3%,96.9% and 95.4% respectively; RSD were 2.7%,2.1%,2.4% and 2.8%(n=9).Conclusion The established analytical method combined with multivariate statistical analysis and content determination is suitable for the quality evaluation of Jiawei Shenghua Fukang granules.
