摘要(Abstract):
目的摸索高纯度抗五步蛇毒血清的制备方法,以降低抗血清治疗毒蛇咬伤时因纯度不高发生的过敏和其他不良反应。方法从免疫后的马血浆中先提取IgG,然后酶解,通过疏水层析得到F(ab′)2片段,以SDS-PAGE电泳确定纯度,以小鼠体外中和试验确定F(ab′)2活性片段的中和能力。结果制备的抗五步蛇毒血清F(ab′)2冻干品纯度质量分数为91.3%,中和五步蛇毒干粉质量比为15∶1。结论采用新方法制备的F(ab′)2活性片段纯度质量分数能从70%提高到90%以上。
Objective To grope a new approach that improve the purity of Agkistrodon acutus antivenin which can reduce allergic response or side effect during the treatment of the patient bitted by the title snake by using low content antivenin.Methods IgG was extracted from the horse plasma immunized by the venom of Agkistrodon acutus,then IgG was subjected to pepsin-digest,ammonium sulfate precipitate and chromatograph on hydrophobic column to obtain the F(ab′)2 active fragment.The purification of antivenin was performed by SDS-PAGE,and the protective test in vitro was performed in mice.Results The result of electrophoresis strip revealed that the purity of F(ab′)2 active fragment was 91.3%.The obtained antivenin could efficiently neutralize the lyophilized Agkistrodon acutus venom with the amount ratio of 15∶1.Conclusions The purity of F(ab′)2 fragment by the new method is improved from 70% to 90%.
