摘要(Abstract):
目的研制一个全新的 2 0 0L不锈钢固定化青霉素酰化酶反应器并在中试中应用。方法在罐内壁上安装三个盛酶柱 ,每根酶柱装入湿纤维状固定化酶 1 5kg,酶活力 76 2IU·g-1(以湿品为基础计算 )。以HBO3 作缓冲液 ,底物青霉素G钾的质量浓度为 80 g·L-1。每批投料 12kg,每批裂解反应 90min。结果 15批之后 ,剩余酶活力 6 85IU·g-1(以湿品为基础计算 ) ,6 APA收率平均达 89 8%。结论 6 APA生产中使用新的固定化青霉素酰化酶反应器将提高 6 APA的收率 ,减少固定化青霉素酰化酶的损耗。
Objective To study on and use a new 200-liter stainless steel immobilized penicillin acylase (IPA) reactor in a pilot plant.Methods Against the reactor's wall,three columns were installed,in each of which 1.5 kg of wet fiberized IPA with 762 IU·g -1 of activity calculated on wet basis was charged.Dissolved 12 kg of penicillin G potassium (PenGK)was dissolved in a HBO 3-NaBO 3 buffer solution to make the concentration of the solution to 80 g·L -1.Then PenGK was split to 6-APA for a period of the fixed time 90 min.Results The residual activities of IPA were 685 IU·g -1 on wet after 15 batches.The yield of 6-APA reached 89.8% on an average.Conclusions The usage of the new immobilized penicillin acylase reactor in the 6-APA production will result in a higher yield of 6-APA and lower loss of immobilized penicillin acylase.
