石龙清血颗粒的GC指纹图谱及含量测定

摘要(Abstract):

目的 建立石龙清血颗粒的GC指纹图谱,同时对其中4种挥发性成分进行含量测定。方法 采用DB-5毛细管色谱柱(30 mm×0.32 mm, 0.5μm),程序升温,流量为1 mL·min~(-1),载气为高纯氮气,FID检测器温度为280℃,气化室温度为270℃,分流比为1∶10,以龙脑为参照物峰,对其进行方法学验证,建立10批药品的GC指纹图谱,并采用中药色谱指纹图谱相似度评价系统(2012版)进行相似度评价,同时测定药品中4种成分的含量。结果 建立了石龙清血颗粒的GC指纹图谱,10批药品的相似度在0.979~0.999,共标定出22个共有峰,并指认出其中4个色谱峰,定量分析条件方法学考察结果良好,10批药品中α-蒎烯、龙脑、阿魏酸、莪术醇的平均含量分别为0.013 1、0.018 4、0.087 0、0.133 5 mg·g~(-1)。结论 建立的石龙清血颗粒GC指纹图谱及4种挥发性成分的含量测定方法,操作简便、准确可靠,可为石龙清血颗粒的质量控制提供参考。
Objective To establish a GC fingerprint of Shilong Qingxue Granules and simultaneously determinate 4 volatile components.Methods The GC separation was achieved on a DB-5(30 mm×0.32 mm, 0.5 μm) column with a temperature program at the rate of 1 mL·min~(-1).High purity nitrogen was used as carrier gas.Flame-ionization detector temperature was 280 ℃ and in injection port temperature was 270 ℃,the split ratio was 1∶10.Borneol was used as the reference peak, and methodological verification was carried out to establish GC fingerprint of 10 batches of drugs.Similarity Evaluation System for Chromatographic Fingerprint of TCM(2012 edition) was used to evaluate the fingerprints.At the same time, the contents of 4 components were determined.Results The GC fingerprint of 10 batches of Shilong Qingxue Granules was established.The similarity was between 0.979-0.999,and 22 common peaks was marked.4 Peaks were confirmed.The result of quantitative analysis were good.The average content of α-pinene, borneol, ferulic acid and curcumol in 10 batches of drugs were 0.013 1,0.018 4,0.087 0 mg·g~(-1)and 0.133 5 mg·g~(-1),respectively.Conclusion The established GC fingerprint of Shilong Qingxue Granules and the content determination method for detecting the 4 volatile components were simple, accurate and reliable, which can provide reference for the quality control of Shilong Qingxue Granules.
