穿龙薯蓣皂苷元对小鼠T淋巴细胞CD69、CD25表达影响的体外研究
Study on the effects of diosgenin from dioscorea nipponica on the expression of CD69 and CD25 on T lymphocytes of mice in vitro

摘要(Abstract):

目的研究穿龙薯蓣皂苷元(diosgcnin,Dio)对小鼠脾脏T淋巴细胞CD69、CD25表达的影响,探讨Dio的免疫调节机制。方法以不同浓度Dio与经刀豆蛋白A(Con A)刺激的T淋巴细胞共同培养,用流式细胞术和Western-blot法检测Dio对CD69、CD25表达的影响。结果 Dio质量浓度在3.75~15 mg·L~(-1)内,对CD69、CD25的表达有抑制作用,随药物质量浓度递增抑制作用增强,7.5mg·L~(-1)为最佳抑制浓度,超过此浓度后,抑制作用逐渐减弱。结论 Dio对T淋巴细胞CD69、CD25的表达有抑制作用,即Dio对T淋巴细胞的早期、中期活化均有抑制作用。
Objective To study the effects of diosgcnin( Dio) on expression of CD69,CD25 on spleen T lymphocytes of mouse,and then explore the immune-regulatory mechanism of Dio. Methods Spleen T lymphocytes of mouse were activated by the sword bean protein A( ConA) with Dio at various concentrations. The inhibition of Dio on expression of CD69 and CD25 on T lymphocytes was analyzed by flow cytometry and Western blot. Results Dio at concentrating range of 3. 75-15 mg·L~(-1) could inhibit the expression of CD69 and CD25. With the increase of concentration, the inhibition was enhanced significantly and 7. 5 mg·L~(-1) was the optimal concentration. The inhibition effect will be decreased when the concentration was higher than 7. 5 mg·L~(-1). Conclusions Dio could inhibit the expression of CD69 and CD25 on T lymphocytes, therefore Dio could inhibit the initial-period and mid-period activation of T lymphocytes.
