电泳光散射法测定异丙酚微乳剂的动电电位

摘要(Abstract):

目的采用电泳光散射法测定异丙酚微乳剂的动电电位 ,为微多相药物制剂物理稳定性的考察提供简便、快速、有效的方法。方法制备 3种处方不同、工艺相同的异丙酚微乳剂Ⅰ、Ⅱ及Ⅲ ,以NICOMP3 80 /ZLS系统分别测定其电泳光散射光谱。结果与无外加电场情况相比 ,有外加电场时 ,乳剂Ⅰ、Ⅱ和Ⅲ的光谱谱线向左频移分别为 (2 9 82± 0 1 2 )、(1 2 5 4± 0 44)及 (2 0 1 8± 1 1 2 )Hz,相应动电电位为 (4 5 40± 0 0 4)、(1 9 0 9± 1 1 2 )与 (3 0 72± 0 46)mv。结论 3种微乳剂电泳光散射光谱有显著差别 ,动电电位的大小顺序为乳剂Ⅰ >乳剂Ⅲ >乳剂Ⅱ。
Objective To measure the zeta potentials of the propofol micro emulsion using the electro phoretic light scattering (ELS) and establish a convenient, rapid and effective method of physical stability study for an ultra fine dispersion preparation. Methods Three preparations were named emulsionⅠ,emulsionⅡ and emulsion Ⅲ respectively and had the same procedures but different recipes had been made. Their ELS spectra were obtained by the Nicomp 380/ZLS system. Results Compared with the unapplied electric field, the main peak with the applied electric field showed the left shifting, and the values were (29 82±0 12), (12 54±0 44), and (20 18±1 12)Hz, separately. The corresponding zeta potentials were (45 40±0 04), (19 09±1 12), and (30 72±0 46) mv. Conclusions Three preparations had differences clearly in the ELS spectra. The order for the micro emulsion zeta potentials was emulsionⅠ,emulsion Ⅲ and emulsionⅡ.
