摘要(Abstract):
目的建立同时测定常山胡柚(Citrus Changshan-huyou Y.B.Chang)果汁中柚皮芸香苷、柚皮苷和新橙皮苷含量的方法。方法采用HPLC法。色谱柱Diamnosil C18柱(200 mm×4.6 mm,5μm);检测波长283 nm;流动相为甲醇-水-冰醋酸(体积比为37∶63∶0.2);流速1.0 mL.min-1;柱温为室温。结果柚皮芸香苷、柚皮苷和新橙皮苷质量浓度分别在72~576 mg.L-1、113~904 mg.L-1和53~424 mg.L-1内与峰面积呈良好的线性关系,回归方程分别为A=6.790×103ρ+1.457×104,r=0.999 6(n=6);A=1.267×104ρ-2.366×104,r=0.999 9(n=6)和A=1.110×104ρ-1.472×105,r=0.999 9(n=6)。平均回收率分别为98.3%、98.0%和98.4%。测得样品中柚皮芸香苷、柚皮苷和新橙皮苷含量分别为157、289和172 mg.L-1。结论方法准确、可靠,可为常山胡柚的质量控制提供参考依据。
Objective To establish an HPLC method for simultaneous determination of narirutin,naringin and neohesperidin in Citrus Changshan-huyou Y.B.Chang.Methods The chromatographic method was carried out on a Diamnosil C18 column(200 mm×4.6 mm,5 μm)with a mobile phase of methanol-water-glacial acetic acid(V∶V∶V=37∶63∶0.2).The flow rate was 1.0 mL·min-1 and the column temperature was room temperature.The detection wavelength was set at 283 nm.Results The calibration curve was linear within the range of 72-576 mg·L-1(r=0.999 6)for narirutin,113-904 mg·L-1(r=0.999 9)for naringin,and 53-424 mg·L-1(r=0.999 9)for neohesperidin,respectively.The average recoveries of narirutin,naringin and neohesperidin were 98.3%(RSD=1.53%,n=6),98.0%(RSD=1.90%,n=6),and 98.4%(RSD=1.35%,n=6),respectively.The average contents of narirutin,naringin and neohesperidin in Citrus Changshan-huyou Y.B.Chang were 157,289 and 172 mg·L-1,respectively.Conclusions The method is accurate and reproducible,and can be used for the quality control of Citrus Changshan-huyou Y.B.Chang.
