摘要(Abstract):
目的建立田基黄中槲皮素-7-O-α-L-鼠李糖苷的含量测定方法,为田基黄药材的质量控制和综合开发利用提供一定的科学依据。方法采用HPLC法。色谱柱为Hypersil C18柱(250 mm×4.6 mm,5μm),流动相为体积分数0.04%的乙腈-磷酸水(体积比77∶23),流速为1.0 mL.min-1,检测波长为371 nm,柱温为35℃。结果槲皮素-7-O-α-L-鼠李糖苷质量浓度在3.12~99.84 mg.L-1(r=0.9998,n=6)内与峰面积呈良好的线性关系;平均回收率为99.1%(RSD=1.5%,n=9)。结论该方法简便、快速、精密度好,可用于田基黄的质量控制。
Objective To establish an HPLC method for the determination of quercetin-7-O-α-Lrhamnoside in Hypericum japonicum.Methods The separation was performed on an Hypersil C18 column(250 mm×4.6 mm,5 μm) with the mobile phase consisting of 0.04% acetonitrile-aqueous phosphoric acid(V∶V=77∶23)at the flow rate of 1.0 mL·min-1.The detection wavelength was set at 371 nm and the column temperature was set at 35 ℃.Results The calibration curve was linear over the range of 3.12-99.84 mg·L-1 for linarin(r=0.999 8).The average recovey was 99.1%(RSD=1.5%,n=9).Conclusions The method is simple,accurate and reproducible,which is suitable for the quality control of Hypericum japonicum.
