摘要(Abstract)：
目的建立同时测定舒胆胶囊中大黄酸、大黄素、大黄酚、大黄素甲醚含量的方法,完善现有质量标准。方法采用HPLC法。用Diamonsil C18柱(200 mm×4.6 mm,5μm)分离,用甲醇-水-磷酸(体积比84∶16∶0.1)进行等度洗脱,流速为1.0 mL.min-1,柱温为35℃,检测波长为254 nm。结果大黄酸质量浓度在0.220 0~4.400 mg.L-1内、大黄素质量浓度在0.162 0~3.240 mg.L-1内、大黄酚质量浓度在0.520 8~10.42 mg.L-1内、大黄素甲醚质量浓度在0.109 2~2.184 mg.L-1内与峰面积呈良好的线性关系,相关系数分别为0.999 7、0.999 9、0.999 8、0.999 9;平均回收率分别为100.5%(RSD=0.84%)、100.4%(RSD=0.51%)、100.8%(RSD=0.29%)、99.8%(RSD=0.47%)。结论本方法简便、快捷,结果准确,重复性好,可作为舒胆胶囊的质量控制方法之一。
Objective To establish an HPLC method for simultaneous determination of rhein,emodin,chrysophanol and physcion in Shudan(traditional Chinese medicines)capsules.Methods The chromatographic method was applied on a Diamonsil C18 column(200 mm×4.6 mm,5 μm) with methanol-water-phosphoric acid(V∶V∶V=84∶16∶0.1) as the mobile phase.The flow rate was 1.0 mL· min1 and column temperature was 35 ℃.The detection wavelength was set at 254 nm.Results The calibration curve was linear within the range of 0.220 0-4.400 mg·L-1(r=0.999 7) for rhein,0.162 0-3.240 mg·L1(r=0.999 9) for emodin,0.520 8-10.42 mg·L-1(r=0.999 8) for chrysophanol,0.109 2-2.184 mg·L-1(r=0.999 9) for physcion;the average recoveries were 100.5%(RSD=0.84%) for rhein,100.4%(RSD=0.51%) for emodin,100.8%(RSD=0.29%) for chrysophanol,99.8%(RSD=0.47%) for physcion.Conclusions The method is simple,accurate and reproducible and can be used for the quality control of Shudan capsules.
