HPLC法测定14个地区山楂中羽扇豆醇的含量

摘要(Abstract):

目的建立测定山楂中羽扇豆醇含量的方法。方法采用HPLC法。色谱柱为Diamonsil C18柱(200 mm×4.6 mm,5μm),流动相为甲醇-水(体积比60∶40),流速为1.0 mL.min-1,检测波长为205 nm,柱温为25℃,进样量为10μL。结果羽扇豆醇质量浓度在0.4324.32 mg.L-1内与峰面积呈良好的线性关系,r=0.999 8(n=5),平均回收率为95.5%(RSD=3.2%,n=9)。结论该方法简便,准确,重现性好,可用于山楂药材的质量评价。
Objective To establish an HPLC method for the determination of lupeol in Fructus Crataegi.Methods Chromatography was conducted on a Diamonsil C18 column(200 mm×4.6 mm,5 μm).The mobile phase was methanol-water(V∶V=60∶40)with a flow rate of 1.0 mL·min-1 and the detection wavelength was set at 205 nm,column temperature at 25 ℃.Results The calibration curve was linear in the range of 0.432-4.32 mg·L-1(r=0.999 8).The average recovery was 95.5% and RSD was 3.2%(n=9).Conclusions The method is simple,accurate,reproducible,and can be used to evaluate the quality of Fructus Crataegi.
