摘要(Abstract):
目的筛选长春西汀体外透皮实验中的接受液。方法以不同溶媒系统作为接受液 ,利用Valia-Chien水平扩散池对长春西汀的体外经皮渗透实验做了研究 ,渗透池的有效扩散面积为1 91cm2 ,按规定时间取样并更换全部接受介质 ,用HPLC法测定长春西汀的经皮渗透量。结果根据药物在不同溶媒中的溶解度大小 ,选择了 3种溶媒系统作为接受液进行考察 ,其中 30 %聚乙二醇 -生理盐水作为接受液不能真实的反映药物的经皮渗透 ,它与氮酮合用能抑制其发挥促渗作用 ;30 %乙醇 -水作为接受液 ,渗透曲线及动力学参数均良好 ,其中以 5 %Azone作为促渗剂 ,增渗比为 8 5 8。结论采用大鼠腹部皮肤及Valia-Chien水平扩散池筛选了长春西汀体外透皮实验中的接受液 ,并筛选了最佳促渗剂 ,为进一步开展长春西汀经皮渗透的研究提供了有益参考
Objective To select the receptor solution in the percutaneous experiment of vinpocetine.Methods The penetration rate of vinpocetine through the rat′s skin in various receptor solutions was measured using the Valia-Chien diffusion cell with an effective diffusional area of 1.91cm 2.Samples were withdrawn from the receptor solution at certain time intervals over 10 h and assayed by the HPLC method.Results Three solution systems were selected according to the solubility of vinpocetine.30%PEG-saline could not well reflect the permeation rate because of the combination with Azone.Azone in 30%-ethanol-water had strong effects on the permeation of vinpocetine with the ER value of 8.58.Conclusions The appropriate receptor solution was selected.The results were useful for the further study of the peameation of vinpocetine.
