Labrasol对利巴韦林角膜透过率的影响

摘要(Abstract):

目的研究辛酸癸酸聚乙二醇甘油酯(caprylocaproyl macrogo lglycerides,商品名为Labrasol)对利巴韦林角膜透过率的影响,探讨其在眼用制剂中的应用。方法采用离体角膜渗透技术考察不同质量浓度的Labrasol对利巴韦林兔角膜透过率的影响。结果 Labrasol的质量浓度分别为5、10、15、20、30、50 g.L-1时,使利巴韦林的表观渗透系数分别增加了5.09、7.27、13.14、14.27、20.27、17.68倍,与对照组比均有显著性差异(P<0.01)。结论质量浓度为5~50 g.L-1的Labrasol均能显著性的增加利巴韦林的角膜透过量,且对角膜无刺激性。
Objective To investigate the use of caprylocaproyl macrogolglycerides(Labrasol) in an ocular drug delivery system. Methods The effects of Labrasol on the corneal permeability of ribavirin were investigated in vitro using isolated rabbit corneas. Results In the presence of Labrasol at a concentration between 5 g·L-1 and 50 g·L-1,the maximum increase in the apparent permeability coefficient(Papp)was 5.09-,1.5-,7.27-,13.14-,14.27-,20.27-and 17.68-fold for ribavirin,respectively,compared with the one in the control(P<0.05). Conclusions 5-50 g·L-1 Labrasol have potential clinical benefits in improving the ocular drug delivery of hydrophilic compound ribavirin.
