玉屏风散对紫外线照射的人皮肤成纤维细胞胶原蛋白合成的影响
Effect of Yupingfeng Powder on the collagen synthesis of human skin fibroblasts irradiated by ultraviolet

摘要(Abstract):

目的 探讨玉屏风散(Yupingfeng Powder, YPF)通过核转录因子NF-E2相关因子2/抗氧化反应元件(nuclear factor erythroid 2-related factor 2/antioxidant response element, Nrf2/ARE)信号通路对紫外线照射的人皮肤成纤维细胞胶原蛋白合成的影响。方法 体外培养人皮肤成纤维ESF-1细胞,紫外线照射建立细胞老化模型,MTT法检测各组细胞活力,比色法、蛋白免疫印迹法检测胶原合成。结果 与紫外线组相比,YPF组的细胞活力、羟脯氨酸含量和胶原蛋白含量显著增加,同时也显著增加胶原蛋白I(Collagen I)、胶原蛋白Ⅲ(CollagenⅢ)、金属蛋白酶组织抑制剂-1(metalloproteinase tissue inhibitor-1,TIMP-1)、金属蛋白酶组织抑制剂-2(metalloproteinase tissue inhibitor,TIMP-2)、Nrf2/ARE信号通路蛋白表达,降低基质金属蛋白酶1(matrix metalloproteinase 1,MMP-1)蛋白表达。与UV+YPF-H+siRNA组相比,UV+YPF-H+Nrf2 siRNA组的细胞活力、羟脯氨酸含量和胶原蛋白含量及Nrf2、Collagen I、CollagenⅢ、TIMP-1、TIMP-2蛋白表达降低,MMP-1蛋白表达增加。结论 玉屏风散通过抑制Nrf2/ARE信号通路促进紫外线照射的ESF-1细胞胶原蛋白合成。
Objective To investigate the effect of Yupingfeng Powder(YPF) on the collagen synthesis of human skin fibroblasts exposed to ultraviolet(UV) via the nuclear transcription factor NF-E2 related factor 2/antioxidant reaction element(Nrf2/ARE) signaling pathway. Methods Human skin fibroblast ESF-1 cells were cultured in vitro,and the cell aging model was established by ultraviolet irradiation. Cell viability was detected by MTT assay, and collagen synthesis was detected by colorimetry and Western blotting.Results Compared with the UV group, the cell viability, hydroxyproline content and collagen content of the YPF group were significantly increased. It also significantly increased Collagen I, Collagen Ⅲ, metalloproteinase tissue inhibitor-1, TIMP-1), metalloproteinase tissue inhibitor 2(TIMP-2), Nrf2/ARE signaling pathway protein expression, reduce matrix metalloproteinase 1(matrix metalloproteinase 1, MMP-1) protein expression. Compared with UV+YPF-H+siRNA group, the cell viability, hydroxyproline content and Collagen content, and the protein expression of Nrf2,Collagen I,Collagen Ⅲ,TIMP-1 and TIMP-2 were decreased, and the protein expression of MMP-1 was increased in UV+YPF-H+Nrf2 siRNA group.Conclusion Yupingfeng Powder promotes collagen synthesis in UV-irradiated ESF-1 cells by inhibiting Nrf2/ARE signaling pathway.
