摘要(Abstract):
目的建立紫花苜蓿中考迈斯托醇的含量测定方法。方法采用RP-HPLC法,使用Kromasil C18柱(250 mm×4.6 mm,5μm),流动相为甲醇-体积分数为0.04%磷酸溶液(体积比为60∶40),流速为1.0 mL.min-1,检测波长为345 nm,柱温为25℃。结果考迈斯托醇质量浓度在0.75~22.50mg.L-1内呈良好的线性关系(r=0.999 6,n=5),平均回收率为98.45%,RSD为2.2%(n=6)。结论RP-HPLC法适用于紫花苜蓿中考迈斯托醇含量的测定。
Objective To develop an HPLC method for the determination of coumestrol in Medicago sativa L..Methods A Kromasil C18 column(250 mm×4.6 mm,5 μm)with a solvent system consisting of MeOH-H2O[w(H3PO4)=0.04%](V∶V=60∶40)was used with a flow rate of 1.0 mL·min-1 and the detecting wavelength was set at 345 nm.The column temperature was 25 ℃.Results The coumestrol curve showed good linear correlation within 0.75-22.50 mg·L-1(r=0.999 6).The average recovery of coumestrol was 98.45% with RSD of 2.2%(n=6).Conclusions The method is simple and can be used to determine coumestrol with satisfactory accuracy and reproducibility.
