摘要(Abstract):

目的建立同时测定了哥王中芹菜素和山奈酚含量的方法。方法采用HPLC法。色谱柱为Diamonsil C18柱(250 mm×4.6 mm,5μm),流动相为甲醇-体积分数为0.4%磷酸-四氢呋喃(体积比40∶50∶10),流速为1.0 mL.min-1,检测波长为365 nm,柱温为35℃。结果芹菜素和山奈酚质量浓度分别在3.0060.0 mg.L-1、0.34760.0 mg.L-1、0.3476.94 mg.L-1内与峰面积呈良好的线性关系,平均回收率分别为100.2%(RSD=0.95%)和99.4%(RSD=1.0%)。结论该方法准确,重复性好,适用于了哥王药材的质量控制。
Objective To establish an HPLC method for simultaneous determination of apigenin and kaempferol in Wikstroemia indica(L.)C.A.Mey..Methods The chromatographic method was carried out on a Diamonsil C18 column(250 mm×4.6 mm,5 μm) with the mobile phase consisting of a mixture of methanol-0.4% phosphoric acid-tetrahydrofuran(V∶V∶V=40∶50∶10).The UV detection wavelength was set at 365 nm.The flow rate was 1.0 mL·min-1 and the column temperature was set at 35 ℃.Results The linear ranges were 3.00-60.0 mg·L-1(r=0.999 6) for apigenin and 0.347-6.94 mg·L-1(r=0.999 2) for kaempferol.The average recoveries for apigenin and kaempferol were 100.2% and 99.4% with RSD 0.95% and 1.0%,respectively.Conclusions The method is accurate,sensitive with good reproducibility and can be used for the determination of the contents of apigenin and kaempferol in Wikstroemia indica(L.)C.A.Mey..
