RP-HPLC法测定坎离砂中槲皮素的含量

摘要(Abstract):

目的建立坎离砂中槲皮素含量测定的HPLC法。方法采用Diamonsil C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-体积分数为0.3%磷酸水溶液(体积比33∶67)为流动相,流速为1.0 mL.min-1,检测波长370 nm,柱温25℃,进样量10μL。结果槲皮素质量浓度在6.312 5~101.0 mg.L-1内线性关系良好(r=0.999 5);平均加样回收率为95.1%,RSD为3.3%(n=9);方法精密度RSD为2.4%(n=5)。结论该方法简便、准确、重现性好,为坎离砂及其新剂型改进提供了一种质量控制方法。
Objective To establish an RP-HPLC method for determination of quercetin in Kanlisha(traditional Chinese medicine).Methods Chromatagraphy was conducted on a Diamonsil C18 column(250 mm×4.6 mm,5 μm) at 25 ℃ and 370 nm.The mobile phase was acetonitrile-0.3% phosphoric acid aqueous(V∶V=33∶67) at a flow rate of 1.0 mL·min-1.Results The calibration curve was linear in the range of 6.312 5-101.0 mg·L-1(r=0.999 5);The average recovery was 95.1% and RSD was 3.3%(n=9);The precisions RSD was 2.4%(n=5).Conclusions The method is simple and accurate.It can provide a method of quality assessment for Kanlisha.
