基于Box-Behnken响应面设计结合熵权法优选当归四逆汤挥发油提取工艺

摘要(Abstract):

目的 利用Box-Behnken响应面设计法结合熵权法优选当归四逆汤中当归桂枝挥发油最佳提取工艺。方法 以液料比、浸泡时间、氯化钠溶液加入比例作为考察因素,挥发油中2种脂溶性成分(藁本内酯、桂皮醛)的含量作为考察指标;基于单因素实验优选考察因素的最佳范围,运用Box-Behnken响应面设计法构建三因素三水平试验设计方案,采用熵权法对2种指标进行赋权,以综合评分结果作为提取工艺的评价指标优选最佳工艺条件,并进行工艺验证。结果 当归桂枝挥发油的最佳工艺条件为液料比4.6∶1,浸泡时间1.14 h,氯化钠加入比例为5.7∶1,提取时间为4 h,提取1次,药材粉碎程度为全部通过1号筛。结论 利用Box-Behnken响应面设计法优化当归四逆汤挥发油提取工艺方法稳定,预测性较好,对当归四逆汤挥发油提取工艺研究具有重大的指导意义。
Objective Box-Behnken response surface design method combined with entropy weight method was used to optimize the optimal extraction process of volatile oil from Angelica Sinensis and Cassia Twig in Danggui Sini decoction.Methods The liquid-material ratio, soaking time and the proportion of sodium chloride solution were used as the investigation factors, and the contents of two fat soluble components(ligustilide and cinnamaldehyde) in volatile oil were taken as the investigation indexes.Based on the single factor experiment, the optimal range of factors was optimized.The Box-Behnken response surface design method was used to construct the three factor and three-level experimental design scheme, and the entropy weight method was used to weight the two indexes.The comprehensive score results were used as the evaluation indexes of the extraction process.The optimal process conditions were optimized, and the process was verified.Results The optimal technological conditions of volatile oil from Angelica Sinensis and Cassia Twig were as follows: liquid material ratio was 4.6∶1,soaking time was 1.14 h, sodium chloride addition ratio was 5.7∶1,extraction time was 4 h, extracted once, and the crushing degree of medicinal materials was all through No.1 sieve.Conclusion Box-Behnken response surface design method is stable and has good predictability, which can be used to optimize the volatile oil extraction process of Danggui Sini decoction.It has great guiding significance for the study of volatile oil extraction process of Danggui Sini decoction.
